If you don't use your legs for 4 years+ it's a little hard to stand up and run around... more like impossible.

edit: sounds weird, lemme be specific: a miracle can bring the feeling back to your legs, maybe, I will buy that gladly, but it can't regrow muscle.

We don't know the timeframe for Locke becoming wheelchair-bound, so it may have not been incredibly long since the incident that hurt him.

Unless he was lying when he yelled at the walkabout tour guide, yeah we do. Four years at least.
