All right, I'm gonna cheat and list a few here...
Zelda 3-SNES (After the first time, knowing where stuff is really killed the game, and made it so much shorter and less enjoyable.)
Seaman-DC (It's just not the same the second time.)
Super Monkeyball-Gamecube (The first time I was all "It's monkeys and balls, and rolling, and punching, and *gasp* can't stop must play with the monkies!!!" Now it's just damn cool.)
I'd have to say the original Resident Evil, Castlevania: Symphony of the Night, Skullmonkeys, Darkstalkers, Vandal Hearts, Super Metroid, Blackthorne, and Street Fighter 2.

When I got this game at age 13 or so, I did nothing that entire summer but play this game. I remember the first time fighting Magus, thinking it was going to be the last boss because who could he a greater evil, but not wanting it to be because that would mean it was about a 6 hour RPG.
I'd do anything to be on the edge of my seat again, wondering where my time-traveling adventures would lead me next. What a game. I think its time to play it again.
