If it's your first trip, I'm not sure there's a lot of value in going to another city outside Tokyo - Tokyo itself is huge and extremely varied, and you're not going to see more than a fraction of what the city has to offer in seven days. There are a ton of side trips you can take out of town - Mount Takao is a good one; Hakone is great for hot springs and exploring the countryside; Nikko has stunning scenery and ancient temples; Kawaguchiko has roller-coasters, hikes and brilliant views of Fuji; Kawagoe is a quaint little town which retains its old buildings and layout - all of these are within an hour or two of Tokyo. Do a couple of those instead of going to another city and you'll save quite a lot of money on the rail pass.
If you really insist on going to another city, don't bother with Osaka (it's got a distinct culture from Tokyo but not distinct enough that you'll notice in a two-day whirlwind visit; physically, it's just mini-Tokyo). Try Kyoto (smaller, surrounded by mountains, great temples etc., but crammed with tourists at all times) or, for something a little off the beaten track, Kanazawa (recently linked to Tokyo by bullet train, much more chilled out than Kyoto but with lots of beautiful buildings and scenery).
