I'll second the online Tetris Attack and Pokemon MMO ideas. Everything else I wanted (New 2D Sonic that is good and new 2D Mario) has been happening on the DS.
New 2D fighters that are good would be nice as well, though.

Edit: Fuck! Just thought of a few things I want to see -- Good online Bomberman on XBLA, online Puyo Pop (preferably on XBLA) and a new Battletoads would all be incredible.

Herzog Zwei: I know Technosoft is dead, but somebody could come up with a rip-off game in it's style. Put in on Xbox Live arcade and offer online multiplayer. This game was beyond great!
