Is this just because of your physical comedy thing? I can't remember anything about Ghostbusters that was essentially masculine, and I don't mean that in the "there is no such thing as essential masculinity" way. I could see how, like, This is the End might not work, or a least might not work as well, with ladies.
If anything, I don't think the political landscape would sustain Ghostbusters today regardless of sex. We don't see East Coast eggheads or the EPA as agents of the apocalypse anymore. Well, not as many of us do, anyway.
Women can be funny. Though I know what you're talking about. I've known way way too many 'funny' girls that don't really understand humor. You know the ones, those 400 pounders that like to make poop and dick jokes between talking about the nightmare before Christmas and Death Note.
There are tons of hillarious female comics. The biggest issue is that hollywood usually writes bad token parts for girls and only casts women based off looks (this being an exception i guess).
I'm sure a female cast will be the least of the problems with this movie.

It will probably be a lifetime movie where the bank is about to take away the house that two of the girl's got from the passing of their mother. At rock bottom, two of their friends help them start a ghostbusters franchise. Then hilarity and mischief ensues! And through it all, the bonds of sisterhood and friendship are made even stronger and 1 mean ghost learns a lesson it will never forget!
