I don't have any particular loyalty to any store, and I keep a close eye on the best deals. If there is a limited edition that I must have, I'll order it from Amazon, unless it is Gamestop exclusive. I don't really buy a lot of new games though.
On the very rare occassion that I buy a retail game at full price I go to Best Buy, since I can just grab the game and checkout with minimal fuss or having to hunt down the guy with the keys to lockup. It's one of the few things Best Buy does right!
I just rediscovered Lockerz.com, which has transformed from an annoying freebie site to a only moderately annoying site where you can watch a few videos and get 20-30% off games. I'm testing them out with some Vita stuff.
I work at Target so I normally buy my stuff to use my pathetic discount (other perk I easy access to clearance stuff). Older stuff I usually buy from Amazon

I neglected to mention that NewEgg has started to beat Amazon on preorders, as $48 straight up is far better than $60 and a $10 credit, so they are getting an increased cut.
I don't know if it has been stated but Best Buy is in hard Financial times. Anyway Best Buy with the Reward Zone Member perks you also get special invitations to private shopping events and bonus points on certain items.
For 360 games, I use the internet (Amazon/eBay). Though I think I only have like 5 or 6 Zboz games total.
For everything else, I frequent a couple of local game shops.
I used to love Newegg, but then they opened up a distribution center in Memphis, TN, so now I have to pay sales tax on everything from there, which kills most of their deals.
Kmart is great during the holiday game season when they're throwing out $20 coupons with every major game and you can just ride the coupon chain for weeks.

I usually go to Fry's first, when it comes to new games. They usually sell newly released games for $10 less than msrp, during the first week and they'll occasionally mark-down new games as well
I buy used PC games (and sometime console games) at Half-Price-Books at least once a month.
The last game I'd bought at Best Buy was a discounted Dead Space 2 (CE) for the PC; It was $20. I haven't really bought much from them lately, and the only reason I've gone there recently is because people keep giving me gift cards to the store.
I love how Online is the highest rated one and no one thought of that when making the poll. Amazon is always cheapest with free release date delivery if you have Prime, no tax, and a lot of times you get money back too. If you're like me, you also have the credit card that gives you rewards. It really is a no brainer compared to going to a store.
