Lock-on will fuck you on multi-monster fights. You're better off getting used to how each weapon behaves, including your directional orientation.
You kill big monsters and then get random drops. You use those drops to make better equipment. That's it, really. There are a lot of bullshit quests like collect this and gather that, run these eggs or rocks over here but those are just bullshit. You need to look up the good end game weapons of whatever you choose and then follow the upgrade path so you don't dead end and waste your time. There's no in-game way to know, you have to look it up. Don't worry about armor until you start dying in one or two hits. At that point make the highest armor value set you can with the parts you already have and keep going. Farming for anything but end game armor is a huge, huge waste of time, and no abilities are worth a shit until the end game anyway.
If you just want to make progress, only complete the Key Quests. They're not labeled in-game, you have to look them up. You do the key quests and an urgent quest comes up. You do the urgent and the next tier of quests comes up. You unlock a lot of shit in single player including upgrades to your cat farm and your food buffs.
You can combine materials to make items you need. After gathering some make your cats farm herbs, blue mushrooms, and honey. Herb + blue mushroom = potion, potion + honey = mega potion. That's the most important shit.
Village quests are stupid super retard easy. Guild quests start off normal. It's a big jump so don't try it alone when your weapon is shit. If you call me and I'm around I can help you kill a lot of stuff. John is always playing this stupid game and he's left me in the dust, he might be willing to help if you see him online.

Listen to Mzo talking about being efficient. I tried to get him to do key quests and he spends all his time delivering eggs offline trying to finish every stupid quest for no reason.
I guess my advice for starting brand new is try out the weapons until you find something that feels good. Then just upgrade your weapon as you play, maybe look at the weapon tree online, but don't worry about it too much at first. The most important thing at first is just seeing the monsters and their attacks, and knowing when you can get hits in without getting hit back.
Also, village quests are not stupid easy at first- you won't get 1 shot but it still takes a while with starting gear and you'll spend a lot of time getting smacked around while you figure out monster attack patterns. I didn't realize this was your first MH Corey. I wrote you a reply on the wiiu yesterday but just text me if you see me on and want to play some, I'll do whatever and answer any questions you have.

Mzo is correct if you want to min max. honestly, just max out any weapon you like. Brachy weapon are hella overpowered. i kill g rank uragaan with the brachy sns in like 12 min on average.
If you have an android phone, there is a ap called mh3u dex. has the important stuff. item drop rates, key quests, and monster weaknesses. the other one, mh3u helper, has the weapons trees.
Complete the villiage, you can to get your food buffs and shipping fleet. it goes to high rank. offline high rank is easier than online high ranks, and the armor you can get will last you until around hr7. and you can get most of the rare 7 and rare 8 weapons, which will really destroy just about anything in hr6. And the shipping fleet is basically free money. send them treasure hunting and you can get from 20k to 60k a run.
Its a good idea to make sets for different weapon type. i have a charm gatherer set for mining. a tank set for gunlance. a evade for hammer/SnS/SA,and a awaken set (gigginox X) for kelbi bow.
Trying to make another Tank Set using Steel Uragaan, but the bastard doesnt want to give me rubies/palliums. But thats how it goes.
